  • The aim of the project. The project develops natural, antifungal agents from microbial origin as effective against fungal disease and represents a biotechnological sustainable approach to the efforts for reducing the damage produced by fungal infections, based on the antifungal abilities of some beneficial microbes. Successful use of microorganisms will be enhanced by understanding the mechanism of biological control. To achieve this goal, bacterial strains metabolite producing and fungal strains toxin producing had to be selected; toxins assayed methods for conditioning bacterial metabolites, analysis of interaction between metabolite and fungal strains had to be developed during the project. Biocontrol methods are offering by many advantages, such as: reduce the use of chemical-based fungicides and the risk of developing pathogen resistant to traditional chemicals; they are safer to use; they are more stable than chemical pesticides if stored properly and have lower re-entry interval times and are less phytotoxic.

OBJECTIVES
  • PRESENTATION
    • In recent years public concern over pesticide residues in the environment, foods and feeds has led to a limitation and / or reduction of availability of some chemical fungicide commonly used to control plant pathogens. The use of chemical compounds has failed to control plant diseases due to resistance, environment pollution, and damage to human health. As a matter of fact, alternative methods of controlling these pathogens and diseases are needed. The use of fungal biocontrol agents is becoming an increasingly important alternative to chemicals crop protection against weeds, insects, and diseases in both agriculture and forestry. The success of biocontrol and yield increase depends on the nature of the antagonistic properties and on the mechanisms of action of the organism.
    • Biocontrol mechanisms can be classified as a) direct competition with the target organism; b) antibiosis; c) predation or parasitism of the target organism and d) induced resistance of the host plant.
    • The complex activities to be developed are based on scientific expertise of all partners involved and comprise the following steps: study and analysis of state of art in domain; elaboration of methodology for selection of bacterial strains producing metabolites with antifungal activity; elaboration of methodology for selection of fungal strains producing toxins; methods for toxin level evaluation; screening of bacterial strains to produce antifungal metabolites; screening of fungal strains producing toxins; evaluation and testing of antifungal activity of bacterial metabolites; evaluation of fungal growth inhibition; evaluation of inhibition of toxin production; study the obtainment of an adequate matrix for metabolites conditioning; optimization of metabolites production; evaluation of physical-chemical factors influencing antifungal activity of microbial metabolites (pH, temperature, proteolytic enzymes); stability of conditioning metabolites.
  • GENERAL OBJECTIVES
    • Improvement of Romanian research quality by knowledge and expertise; development and concentration of human and materials resources; and last but not least, creating of a partnership for a long term
    • The increasing of competitively level in research domain, due to stimulation partnerships to solving some complex problems
    • The development of biotechnologies for preservation and consolidation of biological and ecological diversity, that will be reflected in quality of life and economical progress
    • Supporting the development of biotechnological domain and connected domains (such as biology, chemistry, microbiology, biochemistry), and of existing research network that are able to create excellence researches, conforming to international standards
    • Valorification of microbial potential to obtain useful products and viable solutions to existing problems
    • Improvement of Romanian integration by researches-development activities of high complexity and technical level, with final results as compared with international programs
  • SPECIFIC OBJECTIVES
    • Study of mechanism responsible for biological control of fungal diseases by microbial metabolite with antifungal activity
    • Identified of fungal strains and their toxins production;
    • Selection the most efficient and antagonistic strains for biocontrol ability:
    • To optimize culture conditions for the production of the bacterial metabolite in submerged fermentation
    • Influences of incubation conditions on bacteria growth, sporulation, and production of antifungal metabolites
    • Bacterial culture supernatant minimum inhibitory concentration - testing dilution of metabolites against fungal strains
    • Metabolite characterization as regarding some physical-chemical factors (temperature, pH, proteolytic enzyme denaturation)
    • Studies of the possibility to encapsulate the microbial metabolites - testing the proper matrix for encapsulation
    • Studies of conditioned metabolites stability
    • Development of research units as a consequence of specific equipment acquisition to ensure a high technical level comparative to European one;
    • Dissemination of obtained results (scientific communications, project web page)
